LBgard Blood Tubes are direct-draw blood collection tubes pre-filled with a proprietary, reagent that stabilizes cells and cell-free DNA (cfDNA) in whole blood, preserving biological analytes for downstream applications. LBgard Blood Tubes also prevent the release of cellular components that can interfere with analysis, such as cellular genomic DNA and products from hemolysis. Samples collected in LBgard Blood Tubes, when stored at 4°C – 37°C, are stabilized for downstream circulating tumor DNA and cell-free DNA applications for up to 14 days.

Brake settings are centrifuge-dependent. For the first spin, we recommend light braking, which will not disturb the buffy coat. For the second spin, heavy braking is ok.

Yellowing of color and decrease in additive volume are expected over the lifetime of the LBgard Blood Tube. These changes do not affect functional performance of the tube. Performance is validated for any tube used before its expiration date, provided it has been stored within the indicated temperature range.

Yes, gDNA can be extracted from blood collected in LBgard tubes using the QIAamp blood isolation kit (51106). gDNA yield can be improved if necessary by increasing Proteinase K treatment time.

For slide immunofluorescence assays and flow cytometry assays, we recommend a fixation step.

Standard red blood cell lysis protocols.

We recommend the QIAamp CNA kit (55114) from Qiagen. Other kits validated for use with LBgard tubes are the Norgen Plasma Circulating DNA Purification kit (55100), the QIAamp MinElute kit (36912), the MagMax kit (A29319), the Epi proColon plasma quick kit (M5-02-001), and the Roche cobas cfDNA sample preparation kit (07247737190).

  1. Invert LBgard tubes containing whole blood to mix.
  2. Spin at 1900 rcf for 10 minutes. Transfer top plasma layer carefully to an empty conical tube, carefully avoiding the buffy coat and red blood cell layers.
  3. Spin extracted plasma at 4500 rcf for 15 minutes. Transfer the plasma to an empty tube avoiding any remaining pellets.
  4. Repeat centrifugation Step 3, if desired.
